A major attribute of the cellular response to DNA double-strand breaks (DSBs) is the formation of nuclear "foci", produced by the accumulation of DNA repair and signalling proteins in chromatin regions flanking the break site. The histone variant H2AX and the checkpoint factor MDC1 make up the central regulatory components of nuclear focus formation. This picture shows that human cells depleted of MDC1 (right) are defective in nuclear focus formation at sites of DSBs (the cell nucleus is stained blue).
